There's a good reason they don't use standard OSB for floors is not water proof. If you paint the top and not the bottom it will allow mosture in and will just break down the structure from the bottom up. If you have to use OSB, paint both sides and the ends before you lay it, at least it has a chance then.
Or better still sell the OSB and buy flooring grade chip board or exterior grade ply. Then paint.
